Background InformationMammalian BIG2 (BFA-inhibited guanine nucleotide-exchange protein 2) is encoded by the Arfgef2 gene in rat (NCBI Gene ID 296380). The fungal antibiotic Brefeldin A (BFA; Cat. No. 203729 & 500583) causes disassembly of the Golgi complex and ER swelling in mammalian cells by inhibiting three ADP-ribosylation factor (ARF) guanine nucleotide-exchange factors (GEFs), BIG1, BIG2, and GBF1, all of which interact with various ARFs via their respective Sec7 domains. ARF4/5 activation by GBF1 is shown to facilitate BIG1/2 recruitment to the trans-Golgi network (TGN) and endosomes, where BIG1/2 mediate ARF1/3 activation and lead to recruitment of clathrin adaptors. Independent of its Sec7 domain-mediated GEF activity, BIG2 is also known to interact with other cellular proteins and function as a scaffold for assembling multimolecular complexes involved in several cellular processes (PMID 23386609, 23918382, and 25097827). Cat. No. ABS991, Anti-BIG2, is a rabbit antiserum raised against rat BIG2 C-terminal sequence (UniProt Q7TSU1) that is not present in BIG1. Antiserum cross-reactivity toward human (UniProt Q9Y6D5) and murine (UniProt A2A5R2) BIG2 has been confirmed.
