DescriptionRabbit polyclonal antibody supplied as undiluted serum adsorbed against non-phosphorylated immunogen peptide. Recognizes the ~180 kDa VEGFR-1 protein.
BackgroundThe vascular endothelial growth factor receptor-1 (VEGFR-1, approximately 180 kDa) protein is a member of a subfamily of receptor-tyrosine kinases characterized by the presence of seven immunoglobulin-like domains in the extracellular portion of the molecules and a split tyrosine-kinase domain intracellularly. Also known as Flt-1, VEGFR-1 is expressed primarily in endothelial cells but has also been demonstrated in various tumor cells, trophoblast cells, monocytes, and mesangial cells. The receptor is a glycoprotein but glycosylation is not required for activity. Following ligand binding VEGFR-1 undergoes autophosphorylation with the major sites being Tyr-1213 and Tyr-1333. Phosphorylation of Tyr-1213 in turn allows the binding of Nck, SHP-2 and P13K to the VEGFR-1 protein and subsequent signal transduction cascades initiated. Interestingly, ligand binding to VEGFR-1 overexpressed in cell types other than endothelial cells fails to initiate a proliferative response suggesting a unique signal cascade in the endothelial cell.
