DescriptionBioTracker™ Orange Zymosan Phagocytosis Live Cell Probe
OverviewZymosan A is a compound prepared from yeast cell walls and commonly used to induce inflammatory responses, especially phagocytosis. BioTracker™ Orange Zymosan Phagocytosis Live Cell Probe is Zymosan A labeled with an orange acidic pH indicator.

The fluorescence of this indicator is especially enhanced in an acidic environment such as that found in phagosomes or other acidic organelles. SCT203 is therefore a useful probe for the observaton of phagocytosis via its fluorescence increase, which can be only observed after phagocytosis. This probe emits fluorescence when excited by 532 or 514 nm sources, in addition to others such as the 488 laser that are compatible with the excitation range.

Background InformationZymosan A is a compound prepared from yeast cell walls that is frequently used to induce inflammatory responses, including phagocytosis. SCT203 is Zymosan A labeled with AcidiFluor ORANGE, an acid pH indicator. Its fluorescence is enhanced in the acidic environment of the phagosome. BioTracker™ Orange Zymosan Phagocytosis Live Cell Probe is therefore a useful tool for the detection of the acidified conditions that follow phagocytosis.

Applications
ApplicationThis live cell imaging probe is zymosan conjugated to fluorescent orange dye & can be used to detect phagocytosis using fluorescence microscopy or flow cytometry.
Key Applications
  • Cell Based Assays
  • Flow Cytometry
  • Fluorescence
Application NotesThis fluorescently-labeled zymosan-A enables fluorescent imaging of phagocytic cells performing phagocytosis.

The lyophilized probe may be reconstituted in culture medium to 10 to 50 ug/mL, but concentration should be optimized to the user's application.

SCT203 has been used to analyze phagocytosis by fluorescence microscopy or flow cytometry in overnight-cultured RAW264.7 cells after incubation for 1-4h.
