Background InformationTransmembrane and immunoglobulin domain-containing protein 2 (UniProt: Q96BF3; also known as CD28 homolog, Immunoglobulin and proline-rich receptor 1, IGPR-1) is encoded by the TMIGD2 (also known as CD28H, IGPR1, UNQ3059/PRO9879) gene (Gene ID: 126259) in human. IGPR-1 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 23-150), a transmembrane domain (aa 151-171), and a cytoplasmic domain (aa 172-282). It is widely expressed with higher expression observed in epithelial and endothelial cells, including bronchial epithelial cells of lung, breast glandular and lobular epithelia cells. It plays a role in cell-cell interaction, cell migration, and angiogenesis. IGPR-1 is localized to endothelial adherens junctions, and through trans-homophilic dimerization it regulates endothelial cell-cell adhesion and barrier function. This dimerization stimulates phosphorylation of serine 220, which is required for IGPR-1 to regulate endothelial barrier function and angiogenesis. Through interaction with HHLA2, IGPR-1 is reported to co-stimulate T-cells in the context of TCR-mediated activation and enhances T-cell proliferation and cytokine production via an Akt-dependent signaling cascade. IGPR-1 is also phosphorylated on serine 220 by I B kinase β, which leads to its activation. Activated IGPR-1 then stimulates phosphorylation of AMPK, which in turn phosphorylates ULK1 and Beclin-1 and leads to increased levels of LC3-II. Genotoxic agents such as doxorubicin and flow shear stress are also reported to simulate phosphorylation of IGPR-1 at serine 220. (Ref.: Amraei, R., et al. (2020). J. Biol.
