Background InformationProtein citrullination, also known as deamination, is one of the post-translational modifications, which converts arginine into citrulline residues, which results in loss of a positive charge in the protein and affects protein structure and immunogenicity. The process of citrullination is catalyzed by a calcium-dependent peptidylarginine deiminiases (PADs), which are conserved in all vertebrates. All PADs, with the exception of PAD4, are localized in the cytoplasm. PAD4 is located in the nucleus. Citrullination has been reported on histone proteins, myelin basic protein (MBP) and filaggrin in normal physiological processes and on fibrin and vimentin during inflammation and cell death. RG/RGG is shown to be a consensus sequence for PAD4-mediated citrullination and RG/RGG motifs are of frequent occurrence in RNA-binding proteins. Enhanced RGG citrullination has been reported in cells treated with A23187, a calcium ionophore. Citrullination by PAD4 is shown to suppress protein aggregation and sequestration into stress granules and a significant inhibition of aggregation of FUS, EWS, and TAF15 (FET) proteins has also been reported. This polyclonal antibody specifically recognized citrullinated RGG motifs. It detects multiple citrullinated proteins in HEK293T cells transfected with wild-type PAD4, but not in cells transfected with mutant PAD4. (Ref.: Tanikawa, C., et al. (2018).
