Background InformationRetinol-binding protein 3 (UniProt: P12661; also known as Interphotoreceptor retinoid-binding protein, IRBP, Interstitial retinol-binding protein, RBP3, Protein 7S) is encoded by the RBP3 gene (Gene ID: 281443) in bovine. RBP3 is synthesized with a signal peptide (aa 1-17) and a propeptide (aa 18-22) that are subsequently cleaved off to produce the mature form. It is a lipophilic glycoprotein that is secreted by rod and cone photoreceptors and plays a major role in photoreceptor homeostasis as well as retinoid and polyunsaturated fatty acid transport between the neural retina and retinal pigment epithelium. Its expression is observed early in retinal differentiation. RBP3 shuttles 11-cis and all trans retinoids between the retinol isomerase in the pigment epithelium and the visual pigments in the photoreceptor cells of the retina. It can bind many retinoids and polyunsaturated fatty acids and in addition to retinoid binding, it also plays a role in retinal development and homeostasis. RBP3 also displays thiol-dependent antioxidant activity that maintains the delicate redox balance in the normal retina. RBP3 dysregulation has been linked to retinal diseases. Mice lacking RBP3 exhibit severe early and progressive photoreceptor degeneration that is characterized by a reduction in both length and numbers of cone sheaths. In mammals four protein subunits of RBP3 have been described (M1-4), each consisting of about 300 amino acids. Clone mAb5 has been mapped to M2 subunit of RBP3. (Ref.: Sears, AE., et al.
