Biological Information
ImmunogenA 15 amino acid peptide sequence near the C-terminus of rat NKCC2 (Haas et al. 1987; Gamba et al. 1994; Simon et al. 1996; Mount et al. 1995; Igarashi et al. 1995; Mount et al. 1998). The immunogen peptide is available (catalog number AG205) for use in pre-absortion controls.
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
HostRabbit
SpecificityRecognizes rat Sodium, Potassium, Chloride Cotransporter 2 (NKCC2, BSC1, CCC2). The immunogen shows no significant sequence homology with NKCC1 or TSC.

SPECIES REACTIVITIES: The immunogen sequence is 100% conserved in mouse, 87% in human and rabbit. Reactivity with other species has not been confirmed.

Antibody TypePolyclonal Antibody
Entrez Gene Number
Entrez Gene SummaryThe sodium-potassium-chloride cotransporter isoform 2 is kidney-specific and is found on the apical membrane of the thick ascending limb of Henle's loop and the macula densa. It accounts for most of the NaCl resorption with the stoichiometry of 1Na:1K:2Cl and is sensitive to such diuretics as furosemide and bumetanide. Some Bartter-like syndromes result from defects in this gene.
Gene Symbol
  • SLC12A1
  • MGC48843
  • BSC1
  • NKCC2
UniProt Number
UniProt SummaryFUNCTION: SwissProt: Q13621 # Electrically silent transporter system. Mediates sodium and chloride reabsorption. Plays a vital role in the regulation of ionic balance and cell volume.
SIZE: 1099 amino acids; 121341 Da
SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein.
TISSUE SPECIFICITY: Kidney specific.
DISEASE: SwissProt: Q13621 # Defects in SLC12A1 are the cause of Bartter syndrome type 1 (BS1) [MIM:601678]. BS refers to a group of autosomal recessive disorders characterized by impaired salt reabsorption in the thick ascending loop of Henle with pronounced salt wasting, hypokalemic metabolic alkalosis, and varying degrees of hypercalciuria. BS1 is a life-threatening condition beginning in utero, with marked fetal polyuria that leads to polyhydramnios and premature delivery. Another hallmark of BS1 is a marked hypercalciuria and, as a secondary consequence, the development of nephrocalcinosis and osteopenia.
SIMILARITY: SwissProt: Q13621 ## Belongs to the SLC12A transporter family.
