DescriptionAnti-Estrogen Receptor α Antibody
Alternate Names
  • ER-alpha
  • Estradiol receptor
  • Nuclear receptor subfamily 3 group A member 1
Background InformationEstrogen receptor (UniProt: P06211; also known as ER, ER-alpha, Estradiol receptor, Nuclear receptor subfamily 3 group A member 1) is encoded by the Esr1 (also known as Esr, Estr, Nr3a1) gene (Gene ID: 24890) in rat. Estrogen receptor a (ERa) is a steroid hormone receptor that is involved in the regulation of eukaryotic gene expression and affects cellular proliferation and differentiation in target tissues. Its expression has been reported in mammary gland, uterus, ovary, bone, prostrate, testes, and adipose tissue. In the absence of ligand, it is weakly associated with nuclear components and upon hormone binding its affinity is greatly enhanced. Its DNA binding region is localized to amino acids 190-255 (Domain C), which is highly conserved and contains two Cys2Cys2 zinc finger motifs. This region is essential and sufficient for binding of ER to the estrogen response element (ERE). Domain E, the ligand-binding domain (LBD; aa 316-552), is also highly conserved and is responsible for interaction of the receptor with various estrogens and antiestrogens. In ERa, transcription activation function-1 (AF-1) is localized in the amino-terminal A/B domain (aa 1-189) of the receptor and serves as a ligand-independent transcription activation domain. Activation function-2 (AF-2) resides in the hormone-binding domain (aa 316-600) and acts as a ligand-inducible activation domain. ERa can undergo phosphorylation by CDK2/cyclin A and by CK1 that enhances its transcriptional activity. In many breast cancers, ER activation by estrogens is shown to be responsible for enhanced proliferation, which is counteracted by the presence of ERβ that exerts an antiproliferative effect. (Ref. Paterni, I., et al. (2014). Steroids. 90; 13-29).

UniProt SummaryFUNCTION: Nuclear hormone receptor. The steroid hormones and their receptors are involved in the regulation of eukaryotic gene expression and affect cellular proliferation and differentiation in target tissues. Can activate the transcriptional activity of TFF1.

SUBUNIT STRUCTURE: Binds DNA as a homodimer. Can form a heterodimer with ESR2. Interacts with the NCOA3, NCOA5 and NCOA6 coactivators. Interacts with NCOA7; the interaction is ligand-inducible. Interacts with AKAP13, CUEDC2, HEXIM1, KDM5A, PELP1, PHB2, SLC30A9 and SMARD1. Interacts with MUC1; the interaction is stimulated by 7 beta-estradiol (E2) and enhances ERS1-mediated transcription. Interacts with DNTTIP2, FAM120B and UIMC1. Interacts with MLL2. Interacts with ATAD2; the interaction is enhanced by estradiol. Interacts with KIF18A and LDB1. Interacts with RLIM (via its C-terminus). Interacts with MACROD1. Interacts with FOXL2. Interacts with SH2D4A and PLCG. Interacts with SH2D4A; the interaction blocks binding to PLCG and inhibits estrogen-induced cell proliferation. Interacts with DYNLL1. Interacts with CCDC62; the interaction requires estradiol and appears to enhance the transcription of target genes. Interacts with NR2C1; the interaction prevents homodimerization of ESR1 and suppresses its transcriptional activity and cell growth. Interacts with PRMT2. Interacts with PIK3R1 or PIK3R2, SRC and PTK2. Interacts with RBFOX2. Interacts with EP300; the interaction is estrogen-dependent and enhanced by CITED1. Interacts with CITED1; the interaction is estrogen-dependent. Interacts with DYX1C1, MAP1S and UBE1C.
