DescriptionPurified rabbit polyclonal antibody. Recognizes the ETO protein.
BackgroundOne of the most frequent translocations seen in acute myeloid leukemias (AML) is that which occurs between the AML1 locus on chromosome 21 and a second locus on chromosome 8, the so called ETO/MTG8 (eight twenty one) locus. While the exact function of the normal ETO protein is unknown based on the high content of proline and threonine residues, the putative zinc fingers and the TAF110 homology domains, it is most likely that it functions as a transcription factor. The translocation between AML1 and ETO is observed in 40% of M2-subtype AML's . In the translocation the amino terminal end of AML1 becomes fused to an almost full length ETO protein. The resulting chimeric protein has multiple functions among which are the ability to transactivate the bcl-2 gene in U937 cells, and the M-CSF gene. Both of these activating functions are markedly different from the normal function of the fusion protein, which is the repression of transcription. The normal ETO protein may also has important functions on its own based on the observation of ETO transcripts in normal hematopoietic cells of different lineages and the presence of relatively high message levels in the developing brain as well as the observation that AML/ETO heterozygous mice die in midgestation from CNS hemorrhaging.
