Application NotesWestern blot: 1-10 μg/mL using Chemiluminescence technique; COX-2 runs as a 68-72KDa protein in most cells, sometimes a 90kDa post-translationally modified form is seen as well. CaCo-2 line extracts (induced with serum or growth factors) are good positive controls.

ELISA: 1:10,000 - 1:100,000 using 100 ng of immunogen peptide per well.

Optimal working dilutions must be determined by end user.

SpecificityCyclooxygenase-2. Two isoforms of Prostaglandin H synthase are well characterized, namely COX1 and COX2. Both forms of COX proteins are membrane associated heme proteins containing cyclooxygenase and peroxidase activities. These enzymes are targets of NSAID (Non steroidal anti-inflammatory drugs) such as aspirin. COX2 is a 72 kDa protein having 60% homology to COX1. It is induced by cytokines and mitogens and likely plays a role in inflammatory diseases such as rheumatoid arthritis.

There are two isozymes of PTGS: a constitutive PTGS1 and an inducible PTGS2, which differ in their regulation of expression and tissue distribution. This gene encodes PTGS2, which shows 86% - 89% amino acid sequence identity with mouse, rat, sheep, bovine, horse and rabit PTGS2 proteins, respectively. Human PTGS2 is expressed in a limited number of cell types and regulated by specific stimulatory events, suggesting that it is responsible for the prostanoid biosynthesis involved in inflammation and mitogenesis. The expression of this gene is deregulated in epithelial tumors.
