Background InformationContactin-associated protein 1(Caspr) is a single-pass type I membrane protein belonging to the neurexin family and is comprised of one F5/8 type C domain, one fibrinogen C-terminal domain, two EGF-like domains, and four laminin G-like domains. Caspr is well documented for its interaction with contactin, an interaction that is crucial to the transport of Caspr to the plasma membrane, the mylination of glial cells, and the generation of axoglial junctions. Studies also suggest that Caspr may play a regulatory role in the cell surface expression of contactin and its targeting to different axonal domains. Expression of Caspr is largely observed in the brain, mainly in CNS myelinated nerve fibers within paranodal axoglial junctions.

UniProt SummaryFUNCTION: Seems to play a role in the formation of functional distinct domains critical for saltatory conduction of nerve impulses in myelinated nerve fibers. Seems to demarcate the paranodal region of the axo-glial junction. In association with contactin may have a role in the signaling between axons and myelinating glial cells.

SUBUNIT STRUCTURE: Interacts with contactin in cis form.

SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein (Potential).

TISSUE SPECIFICTY: Predominantly expressed in brain. In myelinated nerve fibers of the CNS predominantly found in paranodal axoglial junctions. In unmyelinated nerve fibers of the CNS diffusely distributed along the entire surface. Weak expression is detected in ovary, pancreas, colon, lung, heart, intestine and testis.

DEVELOPMENTAL STAGE: Detected on postnatal day 7 in cerebellum. Follows a caudorostral progession according to the myelination process. Appears to redistribute from the internode to the paranodal region during myelin compaction and maturation. Expression reaches maximal levels between days 14 and 18 and remains at the same levels until adulthood.

SEQUENCE SIMILARITIES: Belongs to the neurexin family.

Contains 2 EGF-like domains.

Contains 1 F5/8 type C domain.

Contains 1 fibrinogen C-terminal domain.

Contains 4 laminin G-like domains.
Molecular Weight~ 220 kDa observed
