Background InformationBromodomain-containing protein 4 (Brd4), or Mitotic chromosome-associated protein (MCAP), is a chromatin reader protein and kinase that recognizes and binds acetylated histones and plays a key role in the transmission of epigenetic memory across cell divisions and transcription regulation. Brd4 remains associated with acetylated chromatin throughout the entire cell cycle and provides epigenetic memory for post mitotic G1 gene transcription by preserving acetylated chromatin status and maintaining high-order chromatin structure. During interphase, Brd4 plays a key role in regulating the transcription of signal-inducible genes by associating with the P-TEFb complex and recruiting it to promoters. Brd4 is widely expressed and localized to the nucleus and acetylated chromatin. Brd4 also plays a large role in many cancers, and through its regulation of transcription, Brd4 is important in multiple cellular processes including DNA damage responses, aging, and development.
