      Background InformationShort transient receptor potential channel 5 (UniProt: Q9UL62; also known as TrpC5, Transient receptor protein 5, TRP-5, hTRP-5, hTRP5) is encoded by the TRPC5 (also known as TRP5) gene (Gene ID: 7224) in human. TrpC5 is a multi-pass membrane protein with six transmembrane domains, four cytoplasmic domains (aa 1-330; 420-437; 492-512; 625-973), three extracellular domains (aa 352-398; 459-470; 534-603). It also has four ankyrin repeats (aa 31-60; 69-97, 98-124, 141-170). It is expressed in brain with fetal brains displaying higher expression. TRP-5 channels are also localized to nonuniform clusters that are preferentially targeted to axonal growth cones before the majority of synapses have formed, where they play a crucial role in growth cone morphology and motility. It forms a receptor-activated non-selective calcium permeant cation channel and be active alone or as a hetero-multimeric assembly with TrpC1, TrpC3, and TrpC4. TrpC-5 homomers exhibit increased Ca2 + permeability and are preferentially targeted to nascent synapses and axonal growth cones, making them ideally situated to mediate synapse formation. Dysregulation of TrpC5 is shown to interrupt Ca2+ homeostasis in cancer cell and promotes tumor progression and chemoresistance. (Ref.: He D-X and Ma, X (2016). Acta Pharmacol. Sinica. 37(1); 19-24)
