      Background InformationMicrotubule-associated protein tau (UniProt: P10636; also known as Neurofibrillary tangle protein Paired helical filament-tau (PHF-tau)) is encoded by the MAPT (also known as MAPTL, MTBT1, TAU) gene (Gene ID: 4137) in human. Tau is a highly abundant protein found predominantly in the nervous system. Its primary role is to stabilize and support microtubule assembly, which are essential for maintaining the structure and function of nerve cells. It is present throughout the brain, particularly in regions involved in memory and cognition. MAPT gene consists of multiple exons that undergo alternative splicing, resulting in the generation of different isoforms that differ in the number and arrangement of repeats in the microtubule-binding domain. The expression of the MAPT transcripts varies and depends on the stage of neuronal maturation and neuron type. Aberrant aggregation and accumulation of tau protein have been linked to several neurodegenerative diseases collectively known as tauopathies. The most common tauopathy is Alzheimer's disease, where hyperphosphorylated tau protein forms neurofibrillary tangles inside neurons. Other tauopathies include frontotemporal dementia, progressive supranuclear palsy, and cortico-basal degeneration. These diseases are characterized by cognitive decline, motor disturbances, and memory loss.
