      Brand Family Chemicon®
      Trade Name
      • Chemicon
      DescriptionAnti-Calcium Transporter 1 Antibody
      Alternate Names
      • CaT-1
      • ECAC2
Background InformationCaT-1 (rat/mouse 727 aa; human 725 aa, chromosome 7q33-q34; ~85 kDa) is also known as ECAC2, CAT-like (CATL) or TRPV6 (transient receptor potential cation channel, subfamily V, member 6) contains a cytoplasmic N-terminus, 6 ankyrin repeat domains, 6 transmembrane domains, an extracellular pore region between TM5-6, and cytoplasmic C-terminus. CaT-1 shows 75% sequence identity to epithelial Ca++ channel (ECaC) protein from rabbit kidney. In rat, CaT-1 transcript (3-Kb) is expressed in duodenum, proximal jejunum cecum and colon but not in kidney; whereas ECaC in rabbit is expressed in apical cell membranes in distal part of nephron (kidney), duodenum, jejunum and placenta.

      Applications
ApplicationAnti-Calcium Transporter 1 Antibody detects level of Calcium Transporter 1 & has been published & validated for use in ELISA & WB.
      Key Applications
      • ELISA
      • Western Blotting
Application NotesWestern blot: 1-10 µg/mL using ECL. The expected size of CaT-1 is ~83 kDa. However, posttranslational modification may alter its size depending upon the species and cell type.
Immunohistochemistry: Not tested. It is recommended that the antibody be tried at 2-20 µg/mL
ELISA: 1:10,000-1:100,000 using 50-100 ng of control peptide/well.
Optimal working dilutions must be determined by end user.
      Biological Information
ImmunogenSynthetic peptide near the C-terminus of rat CaT-1.
      HostRabbit
SpecificityCalcium transporter 1 (CaT-1, ECAC2). The immunogen sequence has no significant homology with CaT-2 or other proteins.
      Species Reactivity
      • Rat
Species Reactivity NoteOther species have not yet been tested. The immunogen sequence is 100% conserved in mouse, 88% in humans and 94% in Cat-1/htrp8A and htrp8b proteins.

      Entrez Gene Number
Entrez Gene SummaryCalcium-permeable channels, such as TRPV6, participate in neurotransmission, muscle contraction, and exocytosis by providing calcium as an intracellular second messenger. Depending on the tissue, transcellular calcium transport may be regulated by vitamin D, parathyroid hormone (PTH; MIM 168450), or calcitonin (CALCA; MIM 114130).[supplied by OMIM]

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: Q9H1D0 # Calcium selective cation channel probably involved in Ca(2+) uptake in various tissues, including Ca(2+) reabsorption in intestine. The channel is activated by low internal calcium level, probably including intracellular calcium store depletion, and the current exhibits an inward rectification. Inactivation includes both, a rapid Ca(2+)-dependent and a slower Ca(2+)-calmodulin- dependent mechanism, the latter may be regulated by phosphorylation. In vitro, is slowly inhibited by Mg(2+) in a voltage-independent manner. Heteromeric assembly with TRPV5 seems to modify channel properties. TRPV5-TRPV6 heteromultimeric concatemers exhibit voltage-dependent gating (By similarity).
SIZE: 725 amino acids; 83210 Da
SUBUNIT: Homotetramer and probably heterotetramer with TRPV5. Interacts with TRPV5. Interacts with S100A10 and probably with the ANAX2-S100A10 heterotetramer. The interaction with S100A10 is required for the trafficking to the plasma membrane. Interacts with BSPRY (By similarity). Interacts with calmodulin.
SUBCELLULAR LOCATION: Cell membrane; Multi-pass membrane protein (By similarity).
TISSUE SPECIFICITY: Expressed at high levels in the gastrointestinal tract, including esophagus, stomach, duodenum, jejunum, ileum and colon, and in pancreas, placenta, prostate and salivary gland. Expressed at moderate levels in liver, kidney and testis. Expressed in locally advanced prostate cancer, metastatic and androgen-insensitive prostatic lesions but not detected in healthy prostate tissue and benign prostatic hyperplasia.
PTM: Glycosylated (By similarity). & Phosphorylation by PRKCA at the calmodulin binding site delays channel inactivation.
SIMILARITY: SwissProt: Q9H1D0 ## Belongs to the transient receptor family. TrpV subfamily. & Contains 5 ANK repeats.
