The encoded protein is found as a homotetramer (MAT I) or a homodimer (MAT III) whereas a third form, MAT II (gamma), is encoded by the MAT2A gene. Mutations in this gene are associated with methionine adenosyltransferase deficiency.

The disease is characterized by fetid breath and dimethylsulfide excretion. Most mutations are transmitted in an autosomal recessive manner, but autosomal dominant inheritance has also been observed. The clinical consequences of hypermethioninemia are poorly understood. In some individuals, hypermethioninemia is apparently benign; in others, association of neurological problems with null mutations has been observed.
