      Alternate Names
      • signal transducer and activator of transcription 5A
Background InformationSTAT (Signal Transducers and Activators of Transcription) are a family of conserved transcription factors that transduce high-fidelity signals for the cytokine family of ligands and receptors as the principle substrates of JAK kinases (1,2). In resting cells, the STATs exist in an inactive state in the cytoplasm until activated. Activators include cytokines, growth factors, and some peptides. These activate JAK kinases, which when activated, bind to the STAT SH2 (Src homology domain-2) domain resulting in phosphorylation of STAT (1). Upon activation, STAT1, -3, -4, -5A, and -5B all form homodimers. STAT1 and STAT2, as well as STAT1 and STAT3 form heterodimers, and several STATs form tetramers, depending on the variables of the activating ligand (1). After dimerization, the STATs accumulate in the nucleus by way of nuclear transport (1). STAT5A and STAT5B appear to be most important to the function of growth hormone and prolactin and play critical roles in milk production (1,2,3). The phosphorylation of Tyrosine 694/699 is a hallmark of activated STAT5.

      Entrez Gene Number
Entrez Gene SummaryThe protein encoded by this gene is a member of the STAT family of transcription factors. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein mediates the signal transduction triggered by various cell ligands, such as IL2, IL4, CSF1, and different growth hormones. It has been shown to be involved in diverse biological processes, such as TCR signaling, apoptosis, adult mammary gland development, and sexual dimorphism of liver gene expression. This gene was found to fuse to retinoic acid receptor-alpha (RARA) gene in a small subset of acute promyelocytic leukemias (APLL). The dysregulation of the signaling pathways mediated by this protein may be the cause of the APLL.

      UniProt Number
UniProt SummaryFUNCTION: Carries out a dual function: signal transduction and activation of transcription. Binds to the GAS element and activates PRL-induced transcription.
SUBUNIT STURCTURE: Forms a homodimer or a heterodimer with a related family member. Binds NR3C1 By similarity. Interacts with NCOA1, NMI and SOCS7.
SUBCELLULAR LOCATION: CytoplasmBy similarity. NucleusBy similarity. Note= Translocated into the nucleus in response to phosphorylation By similarity.
      PTM: Tyrosine phosphorylated By similarity.
INVOLVEMENT IN DISEASE: Defects in STAT5B are the cause of Laron type dwarfism II (LTD2) [MIM:245590]; also known as Laron syndrome type II or Laron syndrome due to a post-receptor defect. The phenotypic features are consistent with growth hormone deficiency in the presence of normal to elevated circulating concentrations of growth hormone, and resistance to hexogeneous hormone therapy.
SEQUENCE SIMILARITIES: Belongs to the transcription factor STAT family.
      Contains 1 SH2 domain.
      Molecular WeightSTAT5A (94 kDa) and STAT5B (92 kDa)
