Several peptides associated with feeding behavior have been reported recently. Orexins (Orexin-A and Orexin-B) are a family of hypothalamic neuropeptides selectively expressed in the hypothalamus (Sakurai et al., 1998). Orexin-A and Orexin-B are derived from the same precursor (Prepro-orexin) by proteolytic cleavage. Prepro-orexin is an 130 aa peptide with a putative 33 aa secretory sequence, a hydrophobic core followed by residues with small polar side chains. The expression was detected in brain and to a small extent in testis (Sakurai et al., 1998; DeLecca et al., 1998). These neuropeptides bind and activate two closely related Orexin receptors--G protein coupled receptors (GPCRs) OX1R and OX2R.

Anti-Orexin-B Antibody detects level of Orexin-B & has been published & validated for use in ELISA & WB.
Key Applications
ELISA
Western Blotting
Application Notes
Western blot: 1-10 μg/mL using Chemiluminescence technique
ELISA: 1:10,000 - 100,000 using 50-100 ng of control peptide per well.
Optimal working dilutions must be determined by end user.
Biological Information
Immunogen
A 13 amino acid peptide mapping near the amino terminus of mouse Orexin-B. The immunogen peptide has 92% similarity with human Orexin-B.
Concentration
Please refer to the Certificate of Analysis for the lot-specific concentration.
Host
Rabbit
Specificity
Mouse/Rat Orexin-B. Reacts with full length Orexin-B. The AB3100 epitope is not found in Orexin-A. Since Orexins-A and Orexin-B are derived from an unprocessed pre-pro orexins (130 aa), the anti-Orexin-B antibodies may also react with pre-pro orexins.
This gene encodes a hypothalamic neuropeptide precusor protein that gives rise to two mature neuropeptides, orexin A and orexin B, by proteolytic processing. Orexin A and orexin B, which bind to orphan G-protein coupled receptors HCRTR1 and HCRTR2, function in the regulation of sleep and arousal. This neuropeptide arrangement may also play a role in feeding behavior, metabolism, and homeostasis.
FUNCTION: SwissProt: O43612 # Neuropeptides that play a significant role in the regulation of food intake and sleep-wakefulness, possibly by coordinating the complex behavioral and physiologic responses of these complementary homeostatic functions. A broader role in the homeostatic regulation of energy metabolism, autonomic function, hormonal balance and the regulation of body fluids, is also suggested. Orexin-A binds to both OX1R and OX2R with a high affinity, whereas orexin-B binds only to OX2R with a similar high affinity. SIZE: 131 amino acids; 13363 Da SUBCELLULAR LOCATION: Rough endoplasmic reticulum (By similarity). Cytoplasmic vesicle (By similarity). Cell junction, synapse (By similarity). Note=Associated with perikaryal rough endoplasmic reticulum as well as cytoplasmic large granular vesicles at synapses (By similarity). TISSUE SPECIFICITY: Abundantly expressed in subthalamic nucleus but undetectable in other brain regions tested (hypothalamus was not tested) and in heart, placenta, lung, liver, skeletal muscle, kidney and pancreas. PTM: Specific enzymatic cleavages at paired basic residues yield the different active peptides. DISEASE: SwissProt: O43612 # Defects in HCRT are a cause of narcolepsy [MIM:161400]. Narcolepsy is a neurological disabling sleep disorder, characterized by excessive daytime sleepiness, sleep fragmentation, symptoms of abnormal rapid-eye-movement (REM) sleep, such as cataplexy, hypnagogic hallucinations, and sleep paralysis. Cataplexy is a sudden loss of muscle tone triggered by emotions, which is the most valuable clinical feature used to diagnose narcolepsy. Human narcolepsy is associated with a deficient orexin system. Orexins are absent and/or greatly diminished in the brain and cerebrospinal fluid (CSF) of most narcoleptic patients. Human narcolepsy is primarily a sporadically occurring disorder but familial clustering has been observed. SIMILARITY: SwissProt: O43612 ## Belongs to the orexin family.
