      Background InformationCARD11 is part of a group of CARD containing proteins, or proteins that contain caspase recruitment domain, belonging to the membrane-associated guanylate kinase (MAGUK) family functioning as molecular scaffolds at specialized locations of the plasma membrane for the production of multiprotein complexes. The CARD domain is a module for protein-binding that helps to regulate the production of CARD-containing proteins into complexes that signal NF-κB. CARD11 is a multidomain adaptor that is necessary for T-cell receptor (TCR) signaling activation of NF-κB as a necessary component of T-cell activation during adaptive immune response. In this pathway, CARD11 undergoes phosphorylation between its PDZ and coiled-coil domains by protein kinase D (PKC) on serine residues 564, 577, and 657 in order to activate NF-κB. Along with immunity, NF-κB is a critical component in the gene activation for pathways such as inflammation and apoptosis.

      SUBUNIT STRUCTURE: Found in a membrane raft complex, at least composed of BCL10, CARD11, DPP4 and IKBKB. CARD11 and BCL10 bind to each other by CARD-CARD interaction. Interacts (via PDZ domain) with DPP4 (via cytoplasmic tail).

      SUBCELLULAR LOCATION: Cytoplasm. Membrane raft. Note: Colocalized with DPP4 in membrane rafts.

      TISSUE SPECIFICITY: Detected in adult peripheral blood leukocytes, thymus, spleen and liver. Also found in promyelocytic leukemia HL-60 cells, chronic myelogenous leukemia K562 cells, Burkitt's lymphoma Raji cells and colorectal adenocarcinoma SW480 cells. Not detected in HeLa S3, Molt-4, A549 and G431 cells.
