Background InformationHIV and related viruses require coreceptors, in addition to CD4, to infect target cells. Some G-protein-coupled receptors including CCR5, CXCR4, CCR3, CCR2b and CCR8 in the chemokine receptor family, and four new human molecules GPR15, STRL33, GPR1 and V28 were recently identified as HIV coreceptors (1,2). Among them, CCR5 is a principle coreceptor for macrophage- and dual-tropic HIV-1 strains fusion of entry of human white blood cells (3,4). The amino-terminal domain and the extracellular loops of CCR5 serve as HIV binding sites. CCR5 messenger RNA is expressed in lymphoid organs and monocytes.

      Entrez Gene Number
Entrez Gene SummaryThis gene encodes a member of the beta chemokine receptor family, which is predicted to be a seven transmembrane protein similar to G protein-coupled receptors. This protein is expressed by T cells and macrophages, and is known to be an important co-receptor for macrophage-tropic virus, including HIV, to enter host cells. Defective alleles of this gene have been associated with the HIV infection resistance. The ligands of this receptor include monocyte chemoattractant protein 2 (MCP-2), macrophage inflammatory protein 1 alpha (MIP-1 alpha), macrophage inflammatory protein 1 beta (MIP-1 beta) and regulated on activation normal T expressed and secreted protein (RANTES). Expression of this gene was also detected in a promyeloblastic cell line, suggesting that this protein may play a role in granulocyte lineage proliferation and differentiation. This gene is located at the chemokine receptor gene cluster region. Two transcript variants encoding the same protein have been found for this gene.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P51681 # Receptor for a C-C type chemokine. Binds to MIP-1-alpha, MIP-1-beta and RANTES and subsequently transduces a signal by increasing the intracellular calcium ions level. May play a role in the control of granulocytic lineage proliferation or differentiation. Acts as a coreceptor (CD4 being the primary receptor) for HIV-1 R5 isolates.
SIZE: 352 amino acids; 40524 Da
SUBUNIT: Interacts with PRAF2. Interacts with HIV-1 surface protein gp120.
SUBCELLULAR LOCATION: Cell membrane; Multi-pass membrane protein.
TISSUE SPECIFICITY: Found in promyelocytic cells.
PTM: Sulfation contributes to the efficiency of HIV-1 entry. & O-glycosylated, but not N-glycosylated.
SIMILARITY: SwissProt: P51681 ## Belongs to the G-protein coupled receptor 1 family.
