      Description
      OverviewAn amyloidophylic dye that specifically stains stacked β sheet aggregates. Useful for staining and visual detection of amyloid deposits. Does not bind to non-polymerized amyloid peptide. Inhibits the accumulation of the scrapie-associated, protease-resistant isoform of protein PrP without affecting the metabolism of the normal isoform. It is also shown to inhibit Pot1 Binding to Telomeric DNA.

      DescriptionAn amyloidophylic dye that specifically stains stacked β sheet aggre-gates. Useful for staining and visual detection of amyloid deposits in nerve tissue. Binds to amyloid fibrils but not to non-polymerized amyloid peptide. Inhibits the accumulation of the scrapie-associated, protease-resistant isoform of protein PrP without affecting the normal isoform possibly through overstabilization of the conformation. It is also shown to inhibit Pot1 Binding to Telomeric DNA.

      Recommended reaction conditionsDissolve 0.5 g Congo Red in 80 ml of absolute ethanol. To this solution add 20 ml of 10% NaCl solution (2 g/20 ml of water). Filter before use. Immerse coverslips containing cells or sections into this solution for 3 min at room temperature. Wash with several exchanges of distilled H2O until the water runs is clear. Dehydrate with ascending alcohol solutions (50%, 70%, 80%, 95% x 2, 100% x 2). Clear with 100% xylene. Mount coverslip onto slide using Permount or other suitable organic mounting medium and view under a microscope. Amyloid deposits will appear red to pink-red. With a polarizing microscope, amyloid deposits show an apple-green birefringence.
