      Background InformationMdmx is a Mdm2-related protein that like Mdm2 has been shown to bind to and inhibit p53, although not as well as Mdm2. This interaction with p53 occurs via a binding domain located in the N-terminal region of the protein. Mdmx has been shown to counteract Mdm2-dependent p53 degradation and to stabilize p53 in its inactive form. Although Mdmx has significant structural similarity to Mdm2 E3 ubiquitin ligase, most studies have not been able to ascribe E3 ligase function to Mdmx.

The MDM4 putative nuclear localization signal, which all Mdm proteins contain, is located in the C-terminal region of the protein. The mRNA is expressed at a high level in thymus and at lower levels in all other tissues tested. MDM4 protein produced by in vitro translation interacts with p53 via a binding domain located in the N-terminal region of the MDM4 protein. MDM4 shows significant structural similarity to p53-binding protein MDM2. Two transcript variants, one protein-coding and the other likely not to be protein-coding, have been found for this gene.       UniProt SummaryFUNCTION: Inhibits p53- and p73-mediated cell cycle arrest and apoptosis by binding its transcriptional activation domain. Inhibits degradation of MDM2. Can reverse MDM2-targeted degradation of p53 while maintaining suppression of p53 transactivation and apoptotic functions.

      SUBUNIT STRUCTURE: Binds to p53, p73 and MDM2.

      SUBCELLULAR LOCATION: Nucleus.

      TISSUE SPECIFICITY: In all tissues tested, with high levels in thymus.

      DOMAIN: Region I is sufficient for binding p53 and inhibiting its G1 arrest and apoptosis functions. It also binds p73. Region II contains most of a central acidic region and a putative C4-type zinc finger. The RING finger domain which coordinates two molecules of zinc mediates the heterooligomerization with MDM2.
