      Background InformationLHFPL tetraspan subfamily member 5 protein (UniProt: Q8TAF8; also known as Lipoma HMGIC fusion partner-like 5 protein, Tetraspan membrane protein of hair cell stereocilia) is encoded by the LHFPL5 (also known as TMHS) gene (Gene ID: 222662) in human. LHFPL5 is a multi-pass membrane protein with three cytoplasmic domains, four transmembrane domains, and two extracellular domains. Its localization to the plasma membrane requires the presence of protocadherin related 15 (PCDH15). LHFPL5 is recognized as a critical component of the mechano-transduction complex in inner-ear hair cells. It is shown to physically interact with and stabilize transmembrane channel-like protein 1 (TMC1). The C-terminal half of LHFPL5 is involved in this interaction. The N-terminal half is involved in its dimerization and interaction with PCDH15. Mutations in LHFPL5 gene are known to cause autosomal recessive deafness that is characterized by non-syndromic sensorineural hearing loss from damage to the neural receptors of the inner ear. Knockout of LHFPL5 in murine models is shown to result in TMC1 reduction in the hair bundles of auditory cells. (Ref.: Yu, X., et al. (2020). Proc, Natl. Acad. Sci. USA. 117(47); 29894-29903).
