      Background InformationGlycoprotein-N-acetylgalactosamine 3-beta-galactosyltransferase 1 (UniProt: Q9NS00; also known as EC:2.4.1.122, B3Gal-T8, Core 1 O-glycan T-synthase, Core 1 UDP-galactose:N-acetylgalactosamine-alpha-R beta 1,3-galactosyltransferase 1, Beta-1,3-galactosyltransferase, Core 1 beta1,3-galactosyltransferase 1, C1GalT1, Core 1 beta3-Gal-T1, C1GalT1) is encoded by the C1GALT1 gene (Gene ID: 56913) in human. C1GalT1 is a single-pass type II membrane, disulfide-linked, homodimeric protein that displays wide expression. Its expression is higher in kidney, heart, placenta, and liver tissue. C1GalT1 is a Glycosyltransferase that generates the core 1 O-glycan Gal-beta1-3GalNAc-alpha1-Ser/Thr (T antigen), which is a precursor for many extended O-glycans in glycoproteins. C1GalT1 is shown to require a dedicated chaperone, cosmc for its proper function. It plays a key role in angiogenesis, thrombopoiesis, and kidney homeostasis development. Loss of C1galt1 is reported to promote development of aggressive form of pancreatic ductal adenocarcinomas and increased metastasis. Knockout of C1GALT1 gene has been linked to increased tumorigenicity and truncation of O-glycosylation on MUC16, which contributes to increased aggressiveness of tumors. C1GalT1 has a short cytoplasmic domain (aa 1-6), a transmembrane domain (aa 7-29), and a long lumenal domain (aa 30-363). (Ref.: Steentoft, C., et al. (2013).
