      DescriptionPurified mouse monoclonal antibody generated by immunizing mice with the specified immunogen and fusing mouse splenocytes with SP2/0 mouse myeloma cells. Recognizes the ~50 kDa PTPase 1B protein.
      BackgroundProtein tyrosine phosphatases (PTPases) make up an expanding family of transmembrane and intracellular proteins, which function to remove phosphate groups from protein phosphoryl-tyrosine residues. PTPase 1B was initially identified as an active intracellular phosphatase from human placenta made up of 321 amino acids. It was subsequently shown to derive from a larger precursor protein containing 435 amino acids. Further examination failed to show alternative splicing or more than a single copy of the PTP1B in the genome. Purification of a 50,000 kDa protein with phosphatase activity from human placenta demonstrated the protein to have an amino acid composition similiar to that for PTPase 1B and N-terminal analysis revealed an identical sequence. PTPases are involved in regulating processes where protein kinases and protein tyrosine phosphorylation are important. Recently it has been demonstrated that expression of PTPase 1B has profound effects on the transformation induced by the human neu oncogene. Based on this and other observations it has been hypothesized that PTPases have the potential to function as tumor suppressor genes.
