      Immunoprecipitation (1-2 µg/sample)
      Paraffin Sections (1-10 µg/ml, neuraminidase pre-treatment required)
Application CommentsRecognizes human PTHLP, but the homology between the human and rat genes suggests that the antibody may also recognize PTHLP from other species. The antibody shows no cross reactivity with human parathyroid hormone. The immunogen, PTHLP (Peptide-1) Cat. Antibody should be titrated for optimal results in individual systems.

DescriptionPurified rabbit polyclonal antibody. Recognizes parathyroid hormone-like protein (PTHLP).
BackgroundParathyroid Hormone-Like Protein (PTHLP; also called Parathyroid Hormone-related Protein or PTHrP) is a polypeptide factor implicated in a severe neoplastic syndrome, Humoral Hypercalcemia of Malignancy, that is most commonly associated with renal cell and squamous cell carcinomas. It has been purified from tumors of patients with this syndrome and also from the conditioned medium of human tumor cell lines. Oligonucleotide probes, based upon the amino-terminal sequence of PTHLP, have been used to clone several cDNAs corresponding to alternatively spliced mRNAs that are both encoded by a single gene. Homogeneous recombinant PTHLP (141 amino acids) as well as synthetic peptides corresponding to the amino-terminal region of the protein have in vitro and in vivo biological activities as expected for a factor involved with calcium homeostasis. Immunohistochemical staining has shown PTHLP to be present in the keratinocyte layer of normal skin as well as in 34 out of 34 squamous or renal carcinomas but at a much lower percentage in adenocarcinomas.
