DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with NS-1 mouse myeloma cells. Recognizes the ~105 kDa Rb protein.
BackgroundCertain genes have been identified whose function appears to be the supression of tumor formation and are therefore classified as tumor suppressor genes. One such gene has been linked to the hereditary cancer retinoblastoma. The retino-blastoma (Rb) gene encodes a protein of 928 amino acids which is found predominantly in the nucleus and has the capability of binding to DNA. The Rb protein may be modified by phosphorylation but while there are several potential sites for N-linked glycosylation, no modification has been detected. The Rb protein has been reported as having apparent molecular weights of ~105 kDa, ~100-114 kDa, and ~102 or ~104 kDa which are antigenically related. The Rb protein has been detect in at least 20 different common laboratory cell lines with the exception of retinoblastoma cells. The malignant trans-formation of cells is caused by inactivation of the gene by mutation but also by inactivation of the Rb protein resulting from binding to the adenovirus E1A protein or SV40 T antigen.
