      Revision05-September-2007 RFH
ApplicationImmunoblotting (0.5-5 µg/ml, see application references)
DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with NS-2 mouse myeloma cells. Recognizes the ~16 kDa p16 protein.
BackgroundVarious cyclin dependent kinases (CDKs) are critical for progression through the G1 phase of the cell cycle, and the enzymatic activity of CDKs is dependent upon specific association with G1 cyclins. The active cyclin/CDK complex can be inhibited by a number of different inhibitor proteins including the p16 product of the Multiple Tumor Suppressor 1 gene (MTS1). p16 was initially identified as a component of cyclin/CDK complexes and shown to specifically inhibit CDK4, thus earning the name p16INK4. Independent studies have demonstrated that the gene encoding p16 is commonly deleted in many different tumor cell lines. Recent evidence implicates the p16 protein in the Retinoblastoma pathway for control of cell growth and proliferation and demonstrates that p16 can function as a tumor suppressor protein to G1-arrest cells. p16 is also known as p16MTS1, and p16CDK4I. The gene maps to human chromosome 9p21 and has been named MTS1 or CDKN2. Although the role of the CDK inhibitor p16 as a cell cycle regulator is well established, other aspects of p16 function and the issue of alterations in p16 as a primary event in tumorigenesis are still under investigation.
