Background InformationFatty acid-binding protein 2, (FABP2) is a fatty acid-binding protein produced by the intestinal tissues of the small intestine and participates in the transport and intracellular internalization and metabolism of fatty acids in the gut. Its expression is localized to the small intestine, especially within the cellular epithelial cells of the jejunum, with much lower levels in the large intestine. Its expression is confined to the cytoplasm and induced by growth factors such as EGF. FABP2 is also known as FABPI or I-FABP. Because of its role in fatty acid uptake and lipid homeostasis, mutations or polymorphisms affecting the function of FABP2 in fatty acid metabolism have been a source of recent research. In particular, an amino acid substitution known as A54T where the G to A polymorphism of codon 54 of the FABP2 gene results in the substation of a Thr in the mutated form instead of an Ala in the normal form. This Thr-codon 54 mutation demonstrates an increased affinity for long-chain fatty acids and is strongly associated with factors associated with metabolic syndrome disease, including insulin resistance, enhanced fatty acid oxidation, elevated triglycerides and type 2 diabetes. Furthermore, FABPs and in particular , FABP2, play an increasingly important role in controlling the lipid signaling cascade and FABP2 is being studied as key regulatory factor of lipid signaling under normal and pathological conditions. EMD-Millipore’s anti-FABP2 mouse monoclonal antibody, clone 9A9B7B3, has been tested in western blot, paraffin embedded immunohistochemical, and flow cytometry studies using human tissues and cell lines. The clone also reacts with FABP2 from NIH-3T3-L1 mouse cells lines by immunocytochemistry.
