Background Information26S proteasome non-ATPase regulatory subunit 9 (UniProt O00233; also known as 26S proteasome regulatory subunit p27, Homolog of rat Bridge 1, Proteasome macropain non-ATPase subunit 9) is encoded by the PSMD9 gene (Gene ID 5715) in human. PSMD9 is a proteasomal assembly chaperone that forms a stable subcomplex with two AAA-ATPases, PSMC3 and PSMC6, in assisting the assembly of the 20S and 19S particles to form the holo complex. PSMD9 contains a PDZ-like domain (a.a. 108-195) that is reported to interact with a short 4- to 7-amino acid linear sequence motifs (SLIMs) at the C-terminal end of its client proteins, including the RNA-binding protein hnRNPA1, the ribosomal protein S14, the growth hormone CSH1, the transcription factor E12, and the IL6 receptor. PSMD9 and hnRNPA1 interaction is found to play a role in IκBα proteasomal degradation, where hnRNPA1 functions as shuttle receptor that recruits IκBα to the proteasome for degradation.
