      BackgroundMTA1, MTA2, and MTA3 (Metastasis-associated gene 3, ACC# AAH53631) are members of a growing novel gene family associated with highly metastatic human carcinomas. MTA3 is expressed in all tissues as both 515 aa (MTA3) and 594 aa (MTA3L) proteins found in both the nucleus and the cytoplasmic compartments. It was identified as an Estrogen receptor (ER) regulated component of the Mi/NuRD histone deacetylase transcriptional repressor. In response to estrogen, MTA3 directly targets Snail, the master regulator of the epithelial to mesenchymal transition (EMT). Regulation of E-cadherin expression due to MTA3 and ER mediated signaling of Snail expression may be an important pathway in human breast tumors. Loss of ER results in decreased MTA3 expression and tamoxifen reduces expression of MTA3 in endometrial cells.
