      Application CommentsEndotoxins: ≤1 EU/µg anti-IGF-IR. This antibody was selected for its ability to block cell surface IGF-I receptor mediated IGF-I and IGF-II bioactivities. The exact concentration of antibody required to neutralize the human cell surface IGF-I receptor mediated bioactivity is dependent on the IGF concentration as well as on the number and types of IGF receptors present on the cell surface (as a function of cell type and culture conditions). A concentration of 11 µg/ml neutralizes ~50-75% of IGF-I receptor-mediated rhIGF-I biological activity in MCF7 cells. This antibody recognizes a conformational epitope. The detection limit for human recombinant IGF-I receptor by immunoblotting is approximately 25 ng/lane. The detection limit for human recombinant IGF-I receptor is ~0.16 ng/well. Based on immunoblots and ELISA, this antibody does not cross-react with human recombinant IGF-I bp1, human recombinant IGF-I and human recombinant IGF-II. Antibody should be titrated for optimal results in individual systems.

      DescriptionProtein G purified mouse monoclonal antibody. Recognizes the ~250-260 kDa IGF-1 receptor protein.
      BackgroundThe insulin growth factor (IGF) family, which includes IGF-I and IGF-II, binds to the IGF receptors (IGFRs) to mediate a number of biological effects. At least six IGF binding proteins (IGFBP-1-6) have also been identified which complex with the IGFs to modulate interactions with the receptors in either a stimulatory or inhibitory manner. Expression of these proteins is regulated during development, and is both tissue and cell specific. IGF-I exhibits potent effects on motor neuron survival, modulates neurotransmitter activity, and reduces apoptosis during normal development and axotomy. In addition, IGF-I serves a neuroprotective role against β-amyloid induced toxicity. It has also been demonstrated that IGF-I, IGF-II, and several IGFBPs are spatiotemporally regulated following hypoxic-ischaemic brain injuries.
