      Alternate Names
      • PRKCA-binding protein
      • Protein interacting with C kinase 1
      • Protein kinase C-alpha-binding protein
Background InformationPRKCA-binding protein (UniProt Q62083; also known as Protein interacting with C kinase 1, Protein kinase C-alpha-binding protein) is encoded by the Pick1 (also known as Prkcabp) gene (Gene ID 18693) in murine species. PICK1 is a scaffolding protein expressed in many tissues, where it regulates trafficking of multiple membrane proteins. The PDZ (PSD-95/Dlg/ZO-1/DHR (Dlg homologous region) domain (a.a. 22-105) mediates PICK1 interaction with proteins containing PDZ-binding motifs, such as PKC and acid sensing ion channels (ASICs), while the lipid-binding AH (arfaptin homology)/BAR (Bin/amphiphysin/Rvs) domain (a.a. 144-357) mediates PICK1 homo-dimerization and membrane localization. Pick1 gene-disruption results in downregulated ASICs current and calcium transients during process of ischemia-reperfusion and protects against primary murine cortical neurons against injury caused by OGD-Rep (oxygen and glucose deprivation followed by reperfusion). PICK1 is also shown to act as a negatively regulator against ASIC1 phosphorylation/activation by PKA in rat pulmonary arterial smooth muscle cells, where PICK1 facilitates calcineurin-mediated ASIC1 dephosphorylation.
