      Background InformationC-X-C motif chemokine 16 (UniProt Q9H2A7; also known as CXC chemokine ligand 16, Scavenger receptor for phosphatidylserine and oxidized low density lipoprotein, Small-inducible cytokine B16, SR-PSOX, Transmembrane chemokine CXCL16) is encoded by the CXCL16 (also known as CXCLG16, SCYB16, SRPSOX, UNQ2759/PRO6714) gene (Gene ID 58191) in human. CXCL16 functions as the ligand for the chemokine receptor CXCR6 and is one of two chemokines known to exist in both transmembrane and soluble forms. CXCL16 is expressed on leukocytes (macrophages, dendritic cells, B-cells, and monocytes), epidermal keratinocytes, bronchial epithelial cells and renal podocytes. CXCL16 mediates the recruitment and adhesion of CXCR6-expressing cells. In addition to leucocyte recruitment and inflammation, CXCR6 and CXCL16 are also reported to be involved in angiogenesis and their expression shows both positive and negative correlations with cancer patients survival. CXCL16 is initially produced with a signal peptide (a.a. 1-29), the removal of which yields the mature protein with a large extracellular region (a.a. 30-205), a trarnsmembrane segment (a.a. 206-226), and a small cytoplasmic tail (a.a. 227-254). Its extracellular region contains a mucin-like stalk and the CXC chemokine domain (a.a. 32-107) that mediates receptor interaction.

      Application NotesFlow Cytometry Analysis: A representative lot detected CXCL16/SR-PSOX surface expression on myeloid antigen presenting cells (APCs), including monocytes, macrophages, CD11c+ myeloid dendritic cells (mDCs), monocyte-derived DCs (MoDCs), while no CXCL16/SR-PSOX surface expression was found on plasmacytoid dendritic cells (pDCs) (Tabata, S., et al. (2005). J. Leukoc. Biol. 77(5):777-786).
      Flow Cytometry Analysis: A representative lot detected CXCL16/SR-PSOX surface expression on human PBMC-derived dendritic cells (DCs) and PMA-treated human acute monocytic leukemia THP-1 cells, as well as the exogenously expressed human PSOX/CXCL16 on transfected COS-7 cells (Shimaoka, T., et al.       Neutralizing Analysis: The F(ab)'2 fragment from a representative lot inhibited CXCL16/SR-PSOX-dependent bacterial phagocytosis by human PBMC-derived dendritic cells (DCs), PMA-treated human acute monocytic leukemia THP-1 cells, as well as human CXCL16/SR-PSOX-expressing COS-7 cells (Shimaoka, T., et al. (2003). J. Immunol. 171(4):1647-1651).
