Background InformationSerine/threonine-protein kinase 24 (EC 2.7.11.1; UniProt Q9Y6E0; also known as Mammalian STE20-like protein kinase 3, MST-3, STE20-like kinase MST3) is encoded by the STK24 (also known as MST3, STK3) gene (Gene ID 8428) in human. STK24 (MST3) is a member of the highly conserved family of Ste20-like kinases. MST3 is regulated by several posttranslational modifcation events. Autophosphorylation of a threonine residue in the N-terminal kinase domain (a.a. 36-286) is reported to result in autoactivation. and the dephosphorylation of a threonine residue in the C-terminal regulatory domain is shown to promote its binding to the scaffolding protein MO25. In addition, MST3 is known to be cleaved by caspase activity into a 12 kDa C-terminal fragment (a.a.326-443) and a kinase domain-containing 36 kDa N-terminal fragment (a.a. 1-325), which then translocates from the cytoplasm to the nucleus. MST3 is expressed ubiquitously in various tissues including the brain, where it plays an important role in the formation and maintenance of dendritic architecture.
