      Background InformationPannexin-1 (UniProt Q96RD7; also known as Innexin) is encoded by the PANX1 (also known as MRS1, PX1) gene (ORF UNQ2529/PRO6028; Gene ID 24145) in human. Pannexin 1 is a high-conductance, voltage-gated channel protein widely expressed in many organs and tissues, including the sensory systems (eye, inner ear, taste buds, and the olfactory epithelium) and both neuronal and non-neuronal cell types. Three pannexin family genes (PANX1-3) have been identified in human and mouse genomes. Pannexin proteins share structural and functional similarities and play important roles in many physiological and pathological processes, such as ATP release, Ca2+ homeostasis, synaptic neurotransmitters release, apoptosis, and immunological response. Pannexin-1 deficiency causes caspase-3 activation and apoptosis induction of hair cells in cochlear spiral limbus (SLM), resulting in hearing loss among Panx1-/- mice. Panx1 is also expressed in the retina, lens and cornea of the eye, where it plays an important role in mediating the crosstalk between immune and neuronal cells. Pannexin-1 channel opening, frequently together with the activation of purinergic receptors by extracellular ATP, is reported to have a great implication in diverse pathological conditions, including ischemia, trauma, HIV infection, inflammation, migraine, and cancer cell growth.
