      Background InformationHistone-lysine N-methyltransferase 2C (UniProt: Q8BRH4; also known as EC:2.1.1.354, Lysine N-methyltransferase 2C, Myeloid/lymphoid or mixed-lineage leukemia protein 3 homolog) is encoded by the Kmt2c (also known as Mll3) gene (Gene ID: 231051) in murine species. MLL 3 is a histone methyltransferase that catalyzes the methyl group transfer from S-adenosyl-L-methionine to the epsilon-amino group of lysine 4 of histone H3 (H3K4). It is a part of chromatin remodeling machinery that predominantly forms H3K4me1 methylation marks at active chromatin sites where transcription and DNA repair take place. It is a DHHC-type zinc finger protein with eight PHD-type zinc fingers, a SET domain (aa 4763-4879), a post-SET domain (aa 4887-4903) and a RING-type zinc finger (aa 343-388). The SET domain interacts with histone H3, but not H2A, H2B, and H4. In adults, MLL-3 is detected in testis, kidney, spleen, and lung and is weakly expressed in brain. It is absent in heart and liver tissue. Mll3 gene is reported to be frequently mutated in cancers and shows a high frequency of loss-of-function mutations, which are spread over the entire protein, including the PHD fingers and SET domain. Mutations in Mll3 gene are reported to cause defects in BRCA1 associated protein 1 (BAP1) recruitment to the enhancers of tumor-suppressor genes, which represses enhancer activity and promotes tumorigenesis. Depletion of MLL3 can enhance cell migration, but does not elevate the proliferation rate of cancer cells. (Ref.: Zheng, J-Y., et al. (2021). Cell Death Dis. 12(4); Article 364).
